Open Device Manager and see what is states about your sound device.
Start--> Control Panel-->System icon-->Hardware tab--->Device Manager
Are any devices under Sound flagged as having a problem?
Perhaps you need to reinstall your sound driver.... or Windows Media Player.
Assuming your running Windows XP.....

Did you uninstall the old sound drivers (the one for the on-board sound device), before installing the new sound card and it's drivers?
The only thing common to no sound on either sound device is the speakers or the Volume and Mute buttons. Did you check those MUTE buttons to insure there were no ticks in the boxes? Also the volume control on the actual speakers as well... check them and insure your speakers are plugged into the sound output jack.
